Description
Induction of cytochromes P450 (CYP450) by xenobiotic chemicals is a common cellular defense mechanism, usually leading to increased detoxification of xenobiotics but sometimes paradoxically to formation of metabolites that are more toxic and carcinogenic. Nifedipine is a dihdropyridine calcium channel modulator used primarily as an anti-anginal agent, as well as in the treatment of hypertension. It has been shown to activate the pregnane X receptor (PXR), a member of the nuclear receptor superfamily of transcription factors, to induce the expression of genes encoding CYP450 enzymes.
