Specific Aims

1.  Isolation and identification of coregulator (CoR) complexes in mouse tissues

Aim 1.1. Improve protocols for generating protein extracts from mouse tissues of liver, adipocytes, uterus and brain for proteomic analyses
Aim 1.2. Isolate and identify coregulator protein complexes from mouse tissues for analysis by mass spec.

2. Profiling post-translational modifications (PTM) of coregulator complexes

Aim 2.1. Identify PTM sites of CoR complexes in HeLa cells
Aim 2.2. Identify in vivo PTM sites of CoR Complexes in mouse tissues

3. Profiling coordinated PTMs of NR and CoRs following physiologic signaling
 
Aim 3.1. Identify PTM sites of ER, PR and SRC1-3 following physiological stimulation in the uterus
Aim 3.2. Identify PTM sites of PPARgamma and LXR, SRC1-3 and PGC-1 in adipocyte cell lines in the presence and absence of the insulin sensitivity drugs for PPARgamma and LXR
Aim 3.3. In vivo Analyses of PTMs
Aim 3.4. Investigator’s ‘Scoring Guide’ for Coregulators
